Bock et al. — mouse visual cortex with physiology
The first volume to link measured physiology to EM wiring: orientation tuning recorded in a living mouse, then the same cells reconstructed.

Bock and colleagues imaged orientation tuning in mouse primary visual cortex by two-photon calcium imaging, then reconstructed the same tissue by serial-section TEM — about 1,500 cell bodies in roughly 12 TB. It established the structure-and-function paradigm that MICrONS later scaled up.
What it is good for. Seeing the origin of the co-registration problem, which is still the hard part of every functional connectomics experiment.
What it does not support. Dense connectivity analysis. Only a fraction of the volume was reconstructed, and by modern standards the segmentation is sparse and manual.
